The objectives of DD4PPI are to obtain innovative inhibitors of p53 protein-protein interactions in a research framework where excellent science is created by combining computational chemistry, organic synthesis, biology and toxicology.
The rational drug design approach for the synthesis of the inhibitors will involve:
1. Lead generation and optimization for drug discovery of p53 protein-protein interaction (PPI) inhibitors;
2. Innovative synthetic methodologies to prepare the small molecule inhibitors based on indole scaffolds;
3. Screening of compounds as p53 activators;
4. Stability studies of the most promising compounds.
The proposed work plan encompasses a multi-disciplinary team with the required expertise to develop novel libraries of indole-based compounds that are dual inhibitors of p53-MDM2/X protein-protein interactions.
